ABSTRACT

Objective:To explore the absorption and transport properties of flavanomarein in the Madin-Darby canine kidney(MDCK) monolayer cell model. Method:Methyl thiazolyl tetrazolium(MTT) assay was used to investigate the toxicity of flavanomarein in MDCK cells. The resistance value of MDCK monolayer cell model was detected by Millicell-ERS-2 cell resistometer. The effects of mass concentration of flavanomarein,administration time,sodium-glucose cotransporter(SGLTs) inhibitor and glucose transporter 2(GLUT2) inhibitor on the transmembrane transport of flavanomarein were investigated. The concentration of flavanomarein was determined by UPLC-MS/MS, and the apparent permeability coefficient(Papp) and the efflux ratio(ER) were calculated. Result:When the concentration of flavanomarein was 5.625-120 mg·L-1, there was no significant toxic effect on MDCK cells. The transport of flavanomarein in MDCK monolayer cell model was time-dependent and concentration-dependent. The Papp values of flavanomarein were basically between 1×10-6 cm·s-1 to 10×10-6 cm·s-1. Compared with the blank group, the phlorizin group significantly reduced the transport of flavanomarein on the MDCK monolayer cell model at 60 min and 90 min. Conclusion:Flavanomarein is a moderately absorbed drug in the intestine, its transmembrane transport mechanism is dominated by passive transport along with active transport. SGLTs may be involved in mediating the transport of flavanomarein on the MDCK monolayer cell model.

ABSTRACT

OBJECTIVE@#To compare the difference between robotic-assisted TKA and traditional TKA by Meta-annlysis in order to determine whether robotic-assisted TKA can provide better lower limb force line and clinical prognosis.@*METHODS@#Computerized searches of Embase, Pubmed, Web of Science and CNKI, Wanfang and VIP databases were conducted until November 2018 to find out the difference between robotic assisted TKA and traditional TKA. After screening, quality evaluation and data extraction according to inclusion and exclusion criteria, Revman 5.3 software was used for meta-analysis of literature data.@*RESULTS@#Six clinical controlled studies were included, 253 in robot-assisted TKA group and 231 in traditional TKA group. Meta-analysis showed that the incidence of coronal force line angle[WMD=-1.00, 95%CI(-1.66, -0.35), =0.003], coronal force line inversion or valgus>3°[RR=0.04, 95% CI(0.01, 0.13), =0.000 01] in robot-assisted TKA group was significantly better than that in traditional TKA group, but there were no statistical differences between two groups in range of knee joint motion[WMD=0.06, 95%CI(-5.43, 5.55)], =0.98], anterior position tibial angle[WMD=-0.19, 95% CI(-0.81, 0.43), 95%, =0.55] and lateral tibial angle[WMD=-1.37, 95%CI(-3.73, 0.99), =0.25], anterior position femoral angle[WMD=0.30, 95%CI(-1.37, 1.96), =0.72] and lateral femoral angle[WMD=-0.93, 95%CI(-1.98, 0.013), =0.08], the incidence of complications[RR=0.84, 95%CI(0.45, 1.58), =0.60]. The operation time of robot-cassisted TKA group was longer than that of traditional TKA group[WMD=14.28, 95%CI(0.79, 27.77), =0.04].@*CONCLUSIONS@#The application of robotic assistant system in TKA surgery can significantly improve the accuracy of prosthesis fixation, better reconstruct the lower limb line of force, and has the potential advantages of reducing postoperative pain and promoting the recovery of knee joint function. Due to the limitation of the quantity and quality of the included literature, a high-quality randomized controlled study with long-term follow-up is still needed in the future to support the conclusions of this paper.

ABSTRACT

Objective To investigate the anti-inflammatory effect of Cucurbitacin B on the inflammatory reaction of murine alveolar macrophages induced by lipopolysaccharide and its molecular mechanism. Methods The alveolar macrophages were randomly divided into five groups: blank group, LPS group, different concentrations of Cucurbitacin B (1μM, 2.5μM, 5μM)+) +LPS groups. All group cells were pretreated with different concentrations for 2 h, and followed by 1μg/mL LPS. The protein level of TNF-α, IL-1β, IL-6 and PGE2 were detected by ELISA assay, and the NO was detected by Griess methods, and its upstream protein i-NOS,COX-2, Nrf2 and HO-1 were detected by Western-blot. Results The secretion of TNF-α, IL-1β, IL-6 and PGE2 in LPS group were increased significantly than blank groups, while Cucurbitacin B suppressed its protein level inadose-dependent manner. The nuclear erythroid related factor 2 and antioxidant gene were activated while Cucurbitacin B was added into alveolar macrophages in adose-dependent manner. Conclusion Cucurbitacin B can inhibit the extracellar secretion of inflammatory cytokines production, and its molecular mechanism could be due to the promoting Nrf2 translocation to the nucleus, activating the expression of HO-1 gene, reducing the expression of iNOS and COX-2, and thereby alleviating the inflammatory reaction.

ABSTRACT

Objective: To investigate the feasibility and safety of drug-eluting stenting for Wingspan in-stent restenosis Methods: Firebird drug-eluting stenting was used to treat two patients with Wingspan in-stent restenosis (localized restenosis >70% at vertebral artery V4 segment). One of them was a symptomatic case, and the other had contralateral vertebral artery occlusion and bilateral posterior communicating artery hypoplasia. The stents passed through the restenotic site, and they were released under the pressure of 911.93 to 1013.25 kPa, maintaining for 15 seconds. Results: All the stents were successfully released in place. Angiography showed that there was no significant residual stenosis immediately after the procedure. There was no discomfort in patients after the procedure. The patient's preoperative symptom of repeated vertigo attacks was disappeared. No clinical ischemic event occurred in the two patients after following up for more than 6 months. Six months after the restenting, DSA re-examination showed that there was no significant restenosis in one patient. Conclusion: Using drug-eluting stents for in-stent restenosis after Wingspan stenting is technically safe and feasible. However, selection criteria of patients and efficacy of a larger sample study will be needed.

ABSTRACT

<p><b>OBJECTIVE</b>To identify the susceptibility genes of type 2 diabetes in Chinese Han population.</p><p><b>METHODS</b>Single nucleotide polymorphism (SNP) discovery, genotyping and haplotype construction were performed in 30 candidate genes. Case-control study were carried out in a population-based sample and confirmed by the transmission disequilibrium test (TDT) analysis in 77 trio pedigrees. The effects of the SNP rs5210 on gene expression were studied by reporter gene technique.</p><p><b>RESULTS</b>The case-control studies showed that several SNPs on KCNJ11 gene was associated with type 2 diabetes in Chinese Han population, in which the allele frequency of SNP rs5219, the genotype frequency of rs5210, rs2285676 and rs5219, and the frequency of haplotype GA combined of the rs5219 and rs5215 showed significant difference between these two groups (P < 0.05). In addition, TDT test also showed statistical significance on this haplotype GA (P < 0. 05). The reporter gene assay showed that the effect on gene expression was significantly different between two alleles of rs5210 (P < 0.05).</p><p><b>CONCLUSION</b>KCNJII gene is one of the susceptibility genes of type 2 diabetes in Chinese Han population.</p>
